The endorsement came after Lindsey Graham, her brother and longtime senator, died suddenly last week.
Trump announced his support on social media following a meeting with Darline Graham at the White House.
He urged her to enter the Republican primary, stating it would honor her brother's legacy.
"I asked Darline, for the Good of our Nation, to run for the U. S.
Senate in the Special Republican Primary," Trump wrote. He added that she has his "Complete and Total Endorsement" and concluded with "RUN, DARLINE, RUN!"

Darline Graham was sworn into office earlier this week after being appointed by Republican Governor Henry McMaster.
Prior to her appointment, she served as a commissioner at the South Carolina Commission for the Blind and had never held elected office, though she frequently accompanied her brother at campaign events.
Lindsey Graham died at age 71 due to an aortic dissection, according to the District of Columbia medical examiner.
Funeral services are scheduled for July 28 in Washington and July 29 in South Carolina.
The special Republican primary is set for August 11. The winner will face Democratic nominee Annie Andrews in the general election.
