Paolini advanced after defeating Greece's Maria Sakkari.
She now faces 29th seed Alexandra Eala from the Philippines, aiming to replicate her 2024 run to the final.
During her victory on Centre Court, Paolini was aware of Roger Federer watching from the stands.
"It was difficult to play and stay concentrated knowing that my lifelong idol was in the stands watching me," she said.
"Every now and then I told myself not to think about it and stay focused on the match."
