The field at the par-70 course in Fort Worth is now reduced to 132 players.
Denny McCarthy and David Ford also pulled out, with Jimmy Stanger and Brice Garnett entering as replacements.
No official medical reason was given for Koepka's exit.
Reports indicate the withdrawal is a planned rest period after competing for three consecutive weeks, aiming to manage fatigue and reduce burnout risk.
The five-time major champion shot rounds of 63, 69, 66, and 68 at TPC Craig Ranch, finishing at 18-under par.
That result moved him up six spots to 63rd in the FedEx Cup standings, though he remains outside the top 100 in the Official World Golf Rankings.

Defending champion Ben Griffin remains in the field, along with Wyndham Clark, Justin Thomas, Ludvig Aberg, Bob MacIntyre, and Hideki Matsuyama.
Koepka is not eligible for the Memorial Tournament due to his position outside the Aon Swing 5 and Aon Next 10.
