Medina has agreed to a three-year contract with an option to extend for a further season. This marks her first professional move outside Spanish football.
Manchester United's Director of Women's Football, Matt Johnson, expressed strong confidence in Medina's abilities.
"Andrea has all of the technical attributes, character and determination required to thrive at Man Utd," Johnson said.
Johnson noted that the club beat significant competition across Europe to secure the defender.
"We are delighted to add her unique qualities to our group, particularly after significant interest from clubs across Europe," he added.
Despite her young age, Medina has extensive experience at the highest level of European competition.
"Despite being only 22, and still having enormous potential to develop, Andrea already has excellent experience," Johnson said.
"Everyone at the club is excited to help her to achieve her ambitions."

Medina faced Manchester United three times last season in the UEFA Women's Champions League. That experience left a lasting impression on her regarding the club's stature and fanbase.
"The Manchester United has an incredible history, and I'm excited to join a team that is determined to write its own chapter in one of the most important clubs in the world," Medina said.
